Guanosine di- and tri-phosphates in the phosphate metabolism of cerebral tissues promoted by electrical pulses.

چکیده

In a previous communication (Heald, 1957 b) it was shown that the 'phosphoprotein' fraction of cerebral tissues was implicated in the rapid phosphateexchange reactions promoted by applied electrical pulses (Heald, 1956b). It was pointed out that owing to the crudeness of the analytical methods employed the nature of the phosphoprotein could not be determined, more especially since its estimation depended upon a preliminary degradation to inorganic phosphate. It was also considered that since the phospholipid fraction showed no change in specific radioactivity upon the passage of pulses, the phospholipids were not involved in the rapid-exchange reactions which were described. However, it was pointed out by Dr G. H. SloaneStanley and subsequently by Professor J. Folch that treatment of the tissue with trichloroacetic acid to remove phosphates soluble in this extractant before further fractionation would affect the extractability of the phospholipids, some of which might conceivably remain unextracted and be degraded to inorganic phosphate during the analysis for 'phosphoprotein'. For these reasons attempts were made to isolate from cerebral tissues, under conditions less degradative than those previously used, a phosphate fraction or fractions which increased in specific radioactivity when electrical pulses were applied for 10 sec. to tissue which had metabolized radioactive phosphate for a brief period. Such an increase in specific radioactivity under these conditions had previously been found to occur with the phosphoprotein fraction. This has led to the discovery that guanosine diand tri-phosphates are components of cerebral tissue implicated in this type of response. A preliminary account of this work was presented to the Biochemical Society in November 1956 (Heald, 1957 a).

Cerebral Phosphoprotein Promoted by Electrical Impulses

It has been shown (Heald, 1956b) in cerebral slices that duringtherapidbreakdown ofphosphocreatine, brought about by brief passage of electrical pulses (see Heald, 1954), there is an increased exchange of phosphate between the phosphates soluble in trichloroacetic acid and those in the insoluble residue. Thus in cerebral slices which had been allowed to metabolize radioactive inorganic phosphat...

Synthesis of inositol glycan cyclic phosphates.

An efficient synthesis of tri-, tetra-, and pentasaccharide cyclic phosphates 1-5, structurally related to natural inositol phosphate glycans, is reported. The title compounds were assembled by PhSeOTf-promoted glycosylation of the known glucosamine precursor, t-butyldimethylsilyl 2-azido-3,6-di-O-benzyl-2-deoxy-beta-D-glucopyranoside (8) with protected 1-methylthio mono-, di-, and trimannoside...

Interactions of the c-di-GMP riboswitch with its second messenger ligand.

The c-di-GMP [bis-(3'-5')-cyclic dimeric guanosine monophosphate] riboswitch is a macromolecular target in the c-di-GMP second messenger signalling pathway. It regulates many genes related to c-di-GMP metabolism as well as genes involved in bacterial motility, virulence and biofilm formation. The riboswitch makes asymmetric contacts to the bases and phosphate backbone of this symmetric dinucleo...

The Effect of pH on the Liquid-liquid Equilibrium for a System Containing Polyethylene Glycol Di-methyl Ether and Tri-potassium Citrate and its Application for Acetaminophen Separation

In this work liquid-liquid equilibrium for aqueous two phase system composed of polyethylene glycol di-methyl ether and tri-potassium citrate at different medium pH values (6.00, 7.00 and 8.00) and 298.15 K was studied. The obtained results show that two phase area expanded with an increasing of pH values. The performances of the Merchuk and semi-empirical equations were tested in correlating t...

The Relation of Insulin to Phosphate Metabolism *

The early work of Embden, Parnas, and others showed that there was an intimate relation between the metabolism of phosphate and carbohydrate in yeast, microorganisms, and mammalian tissues. Since then the field has been expanded until now it includes a fairly comprehensive scheme. A brief epitomization of this scheme is necessary for the understanding of the possible relations of insulin to pho...
